摘要

目的:建立黄芪健胃膏高效液相色谱(HPLC)特征图谱,提高其质量控制水平。方法:采用Agilent C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,检测波长为230 nm(0~27 min),290 nm(27~40 min);流速:1.0 mL·min-1;柱温:25℃。以芍药苷作为参照峰,测定15批黄芪健胃膏样品,建立对照特征图谱,确定特征峰并采用对照品随行对照定性,阴性对照和单味药材进行峰归属鉴定。结果:建立的HPLC特征图谱,确立了10个共有特征峰,均归属到各药材,并指认了其中7个特征峰。结论:该方法建立的黄芪健胃膏特征图谱专属性强,重复性好,可对该成品同时进行多成分定性分析,可用于该制剂的质量控制。

Abstract

OBJECTIVE To establish the specific chromatograms of Huangqi Jianwei concentrated decoction by HPLC, so as to improve its quality control.METHODS The separation was performed through Agilent C18 column (250 mm×4.6 mm, 5 μm), by gradient elution with the mobile phase consisted of acetonitrile-0.1% phosphoric acid, flow rate at 1.0 mL·min-1, detection wavelength at 230 nm (0-27 min) and 290nm (27-40 min), and the column temperature was kept at 25℃. Using paeoniflorin peak as the reference, the characteristic map was established by 15 batches of Huangqi Jianwei concentrated decoction samples. The characteristic peaks were determined and identified by qualitative, negative control and single herbs.RESULTS Ten chromatographic peaks were determined as common characteristic peaks of the HPLC characteristics and attributed to the herbs. Among them, seven characteristic peaks were identified.CONCLUSION The method to establish the specific chromatogram of Huangqi Jianwei concentrated decoction has high specificity and good repeatability, and can be used for multi-component qualitative analysis and quality control of this product.
